Thanks, Rick. What type of sealant would you use? I need to glue plywood to MDF, and it's a large area. Should I seal all sides of the MDF before assembling, or will the sealant prevent the glue from working?

Not exactly sure what you are doing. I would seal after gluing or the glue won't bond as well unless you sand off the sealant to where you want to glue to. The only problem with a water based glue is the warp and balance of gluing to one side. You know the thing about gluing to both sides of a substrate for balanced construction.

I'm making a sliding shelf (to hold a computer keyboard) for a built-in desk. The drawers that will be replaced are supported by a frame of laminated MDF that screws to the underside of the desktop, so I made the shelf and its frame out of Ultralite.
This MDF doesn't have plastic laminate on it, so the shelf needs to be surfaced with something smooth and durable. There's a piece of 1/4" birch plywood in the garage, which I will glue to the top of the shelf. But that leaves the shelf unbalanced, so I plan to seal the MDF--in the hope that sealant will keep it dimensionally stable.
Will this work?

That's a fairly small piece so it should be ok. It might warp a bit if you use a water based glue. Why not just paint it or laminate both sides with formica. I was originally thinking you were doing something much larger. Seal it right after you laminate with the birch ply.
