I am installing a 1250 cfm dust collector. I would like to know what size duct I should use. Should I use a 6″ main, or is 4″ adequate??

A 4" line would significantly constrict the airflow. It's "adequate" in the sense that it would still work, as long as you keep all but one blast gate closed and use only one machine at a time. But you won't be getting your money's worth out of the DC, and if the line is more than a few feet long, the reduction in airflow compared to a 6" line will be very noticeable.
A 6" main line with 4" drops to the individual machines should be sufficient (or better yet, a 6" main line with 5" drops).
