I’m looking for advice on the best way, regardless of complexity, to attach to plywood edges with machine screws. I want to attach a removeable stiffner brace to the edge of 3/4 Baltic Birch plywood. The attachment will be made with a jig knob on a 1/4-20 machine screw into a threadded insert of some kind, as yet not selected. The design must withstand some rough handling of the plywood set-up.

This is not complex: Select your threaded insert and install it as per the manufacturer's instructions. You could add a bit of CA or epoxy into the drilled hole prior to installing the insert, but that probably isn't necessary. Although it does add some complexity.
